Background of Lysosomal Calcium Ion Channels

Lysosomes are intracellular Ca2+ storage organelles that play an important role in a variety of membrane transport processes. As with the fusion of synaptic vesicles to the plasma membrane, the fusion of lysosomes to other membranes is specifically Ca2+ dependent. In addition, impairment of lysosomal Ca2+ homeostasis and membrane transport has been associated with many human diseases such as lysosomal storage disease (LSD), neurodegeneration, myopathy and cancer. Lysosomal membrane proteins, particularly ion channels, are essential for lysosomal Ca2+ signaling. Due to the intracellular localization of lysosomes and the lack of viable functional assays directly applicable to the natural environment, the role of lysosomal ion channels in Ca2+ signaling is less well understood than that of ion channels in the plasma membrane. The development of biomedical methods has made it possible to monitor lysosomal ion channels and their intracellular Ca2+ signaling.

Direct Monitoring of Ca2+ Levels in the Lysosomal Lumen

(1) Ca2+ Indicators

We select Ca2+ indicators with low sensitivity to pH to monitor lysosomal Ca2+ levels, such as Oregon Green 488, BAPTA-5N .

(2) Small Molecule Fluorescent Ca2+ Indicator Dyes

We used Fura-2 to measure measure the relative lysosomal Ca2+ content, allowing quantitative measurement of cytoplasmic Ca2+ and GCamp or GECO adhering to the cytoplasmic surface of lysosomal membrane proteins.

(3) Genetically Encoded Ca2+ Indicators (GECIs)

We used GECIs to selectively detect lysosomal Ca2+ signals, allowing the study of lysosomal Ca2+ channel activity at high spatial and temporal resolution.
