Lysosomes play a key role in cellular metabolism, endocytosis and synthesis/assembly of hydrolytic enzymes involved in macromolecular digestion. Lysosomes have an acidic inner lumen (pH 4.0-6.0) and contain a variety of proteases that are active at acidic pH. Currently, there are two main types of lysosomal markers, weakly basic amines (DAMP, neutral red, acridine orange) and LysoTracker probes, which selectively accumulate in lysosomes associated with acidic lumen pH. Nevertheless, these compounds still have defects. For example, DAMP is not fluorescent and additional fluorophores must be used to visualize lysosomal staining. In addition, neutral red and acridine orange, which normally stain acidic organelles, are not specific for lysosomes. elevated cell pH during prolonged intracellular accumulation of LysoTrackers may lead to quenching of the fluorochrome in addition to morphological/ physiological changes in lysosomes.

CD BioSciences' lysosomal staining kits use a proprietary lysosomal dye that selectively accumulates in the lysosome through a lysosomal pH gradient. Soluble indicators are hydrophobic compounds that readily penetrate intact living cells and become trapped in the lysosomes upon entry, resulting in bright and specific lysosomal fluorescent staining. Lysosomal scramblers (e.g. chloroquine) are commonly used as positive controls to monitor changes in lysosome number and volume. It is suitable for proliferating and non-proliferating cells, and can be used for both suspension and adherent cells.
