Introduction of Lysosomal Positioning
The way scientists think about lysosomes has changed dramatically in recent years. Lysosomes are now considered key regulators of cellular homeostasis and adaptation to stress. Cells regulate lysosome number and activity in response to a variety of external and internal stimuli. Several protein complexes are associated with the regulation of lysosomal positioning. The distribution of lysosomes within cells is regulated by various stimuli and this regulation is altered under different pathological conditions. Studies have shown that redistribution of lysosomes to the cell periphery is critical for cancer growth, invasion and metastasis. In addition, lysosomal positioning plays an important role in the immune response. Although lysosomes are widely distributed within cells, they can be distinguished spatially based on their location. An emerging and interesting topic is how lysosomal function is influenced by lysosomal positioning. Therefore, adopting the right strategy to assess lysosomal positioning is of great interest for this study.

Our Lysosomal Positioning Assessment Services
Our engineers assessed lysosomal positioning by visual inspection of lysosomal clustering, scoring lysosomes as clustered or unclustered in images of different domains selected in an unbiased manner or in a blinded manner. - We accomplish automated measurement of lysosome clustering by measuring the Clark Aggregation Index (CAI) and the distance of each lysosome from the nucleus, boundary, or center of mass.
- We assess lysosomal positioning by calculating the fraction of total lysosomes (intensity) for each subcellular region of interest.
- We complete the lysosomal positioning assessment by evaluating the peri-Golgi region and calculating the percentage of total lysosomal marker fluorescence intensity in that region.
- We can morphologically analyze lysosomes and their distribution under normal or pathological conditions.
- We quantified lysosomal distribution using the Fiji map, including linear distribution, radial distribution and cumulative intensity distribution.
