Description of Lysosome Isolation Kit (LIK002)

Lysosomes are organelles that are ubiquitous in most eukaryotic cells. They contain many acid hydrolases, which are involved in the degradation of intracellular proteins. Lysosomes also contain lipases, polysaccharases and nucleases. The Lysosome Isolation Kit (LIK002) provides procedures to isolate enriched lysosomal fractions from animal tissues and cultured cells by differential centrifugation followed by density gradient centrifugation and/or calcium precipitation.

Pack Size

The kit is sufficient for the preparation of lysosomes from 25 g of tissue or 20 ml of packed cells.

Applications

Lysosome Isolation Kit has been used:
• in the isolation of cytosol and lysosome.
• to obtain enriched light and heavy lysosomal fractions from cultured cells.
• in the isolation of lysosomes of mouse bone marrow macrophages (BMMs).
• in lysosome purification.

Components

1. Extraction Buffer, 5×100 mL.
2. Optiprep Dilution Buffer, 20×20 mL.
3. Protease Inhibitor Cocktail for use with mammalian cell and tissue extracts, 5 mL.
4. OptiPrep Density Gradient Medium 60% (w/v) solution of iodixanol in water, 100 mL.
5. Neutral Red Reagent, 1 mL.
6. Calcium Chloride Solution 2.5 M calcium chloride solution, 1 mL.
7. Sucrose Solution, 2.3 M 25 mL.

Storage

After receiving the kit, the Protease Inhibitor Cocktail should be stored at -20°C and the OptiPrep Density Gradient Medium should be stored at room temperature. All the other components in this kit should be stored at 2–8°C.

The components are stable for 24 months when stored unopened.
