Background of Lysosomal Proteomics
Lysosomes play an important role in maintaining the normal function of cells, and defects in lysosome function will lead to a series of diseases. Studies have found that lysosomal proteins are also implicated in a wide range of human diseases. Therefore, there has been great interest in understanding the cellular functions of lysosomes. Advances in mass spectrometry and proteomics have laid the necessary foundation to facilitate the study of lysosomes and have become powerful tools for the identification of new lysosomal proteins. To date, it has been identified that the lysosomal membrane contains more than 100 proteins that provide stability, facilitate small molecule transport, and are involved in interactions with other organelles/cellular compartments. Lysosomal lumen contains more than 70 different acid hydrolases, which can be divided into proteases, phosphatases, sulfatase, lipase, carbohydrase, glycosidase and nuclease, of which more than 120 proteins are associated with the lysosomal membrane interaction.
